چکیده

Summary Ribosome biogenesis is a fundamental multi-step cellular process that culminates in the formation of ribosomal subunits, whose production and modification are regulated by numerous factors. In this study, we analyze physiologic prokaryotic ribosome isolating bona fide pre-50S subunits from an Escherichia coli strain with factor ObgE, affinity tagged at its native gene locus. Our integrative structural approach reveals network interacting factors consisting YjgA, RluD, RsfS, ObgE on immature subunit. addition, our study provides mechanistic insight into how GTPase concert other factors, facilitates maturation 50S functional core both conserved divergent evolutionary features between prokaryotes eukaryotes.

Analysis of ribosome biogenesis factor-modules in yeast cells depleted from pre-ribosomes

Formation of eukaryotic ribosomes requires more than 150 biogenesis factors which transiently interact with the nascent ribosomal subunits. Previously, many pre-ribosomal intermediates could be distinguished by their protein composition and rRNA precursor (pre-rRNA) content. We purified complexes of ribosome biogenesis factors from yeast cells in which de novo synthesis of rRNA precursors was d...

Functional specialization of ribosomes?

Ribosomes are highly conserved macromolecular machines that are responsible for protein synthesis in all living organisms. Work published in the past year has shown that changes to the ribosome core can affect the mechanism of translation initiation that is favored in the cell, which potentially leads to specific changes in the relative efficiencies with which different proteins are made. Here,...

Structural Snapshots of Actively Translating Human Ribosomes

Macromolecular machines, such as the ribosome, undergo large-scale conformational changes during their functional cycles. Although their mode of action is often compared to that of mechanical machines, a crucial difference is that, at the molecular dimension, thermodynamic effects dominate functional cycles, with proteins fluctuating stochastically between functional states defined by energetic...
